Furin is a protein that is expressed in the majority of vertebrates, its function is to activate different protein precursors. Earlier research has shown the crucial role that the sodium and calcium Ions play in the activation of the Furin protein. Nevertheless, the energetic and structural aspects that determine its function have been barely studied. To fully comprehend this particular phenomenon might prove crucial in finding and developing new inhibitors with pharmaceutical potential. This is not an easy task; it requires decoupling the different energetic elements. To solve this problem, we propose the use of Free Energy Perturbations; this will allow us to find the role the ions play in the activation process. Through this analysis we found the energetic properties of different conformations of Furin, which helps to understand the importance for function and stability of different components of the protein.
